Depew attended Trinity College, and served as organist and choirmaster of the First Presbyterian Church in Detroit, Michigan. Around 1904, he moved to New York City, where he played the organ at the John Wanamaker Auditorium; Plymouth Church, Brooklyn; St. Nicholas Collegiate Church; and the Capitol and Strand Theaters.
